AIM,REQUIREMENTS,PROCEDURE

• Aim:~
• To study the morphology and motility of bacteria in the given suspension
• Requirements:~
• Cavity slide, cover slip, Vaseline, microscope and the bacterial suspension to be examined

PROCEDURE

• Apply vasiline on the corners of a clean cover slip.
• Using a sterile loop, a loop full of the given suspension is placed on the corner of the cover slip.
• A cavity slide is inverted over the cover slip so that the drop of suspension is in the centre of the cavity

• The slid is quickly and care fully turned over so that the cover slip on the top with the drop hanging into cavity.
• The microscope is adjusted for reduced light by lowering the condenser and using the concave mirror

• The edge of the drop is focused under low power.
• The microscope in then turned to high power to observe the morphology of the bacteria.
• The shape and motility of the bacteria in the given suspension are observed.
